Description:
Leflunomide is a conventional disease-modifying antirheumatic drug (DMARD). It acts by converting to its active metabolite called teriflunomide. This metabolite acts by inhibiting the mitochondrial enzyme dihydro-orotate dehydrogenase (DHODH). This restriction halts the proliferation of activated lymphocytes promoting anti-inflammatory and immunomodulatory effects. Leflunomide has also demonstrated clinical improvements in the signs and symptoms of RA that were achieved as early as 4 weeks after initiation of therapy and were maintained for up to 5 years.
